The Effects of Epidermal Growth Factor and Insulin on c-fos mRNA Proto-Oncogene Expression in the Human Endometrial Cancer Cell Line HEC-1-A


Abstract
Growth factors stimulate cell proliferation through binding to specific high affinity receptors.
Signaling pathway that mediate the normal functions of growth factors are commonly subverted in cancer. Proto-oncogenes are normal cellular genes whose alteration by mutation or deregulation has been implicated in the process of tumorigenesis and
they
play a role for control of normal cellular growth and differentiation.
Epidermal growth factor is mitogenic in moderately differentiated endometrial adenocarcinoma cell line HEC-1-A. This studies were performed to determine the effects of EGF and insulin on c-fos mRNA expression, and whether EGF potentiates insulin
effects
and insulin need new protein synthesis in induction of c-fos mRNA expression in the human endometrial cancer cell line HEC-1-A.
The results obtained are as follows:
1. The maximum expression of c-fos mRNA transcripts were induced at 1 hour stimulation of EGF and insulin in HEC-1-A cells.
2. EGF and insulin induced c-fos mRNA expression as dose dependent manner in HEC-1-A cells.
3. Anisomycin potentiated insulin stimulation of c-fos mRNA expression in HEC-1-A cells.
4. EGF augmented insulin stimulation of c-fos mRNA expression in HEC-1-A cells.
The present studies suggested that insulin may induce c-fos mRNA proto-oncogene expression and EGF may potentiate insulin stimulation of c-fos mRNA transcriptions through protein synthesis independent mechanism.
